RESUMO

The South Orkney Islands (SOI) is a poorly studied Antarctic archipelago located in the Scotia Arc. In this study, we added 25 Demospongiae species to the SOI known sponge richness, which was previously represented by only 16 species. Of these, 22 species represent new records for SOI region, although they were previously recorded from other Antarctic sectors. Samples were collected during two Argentinean Antarctic Research Cruises onboard RV Puerto Deseado. The most frequently collected species were Artemisina apollinis, Iophon gaussi, I. unicorne, Lissodendoryx (E.) ramilobosa, Myxodoryx hanitschi and Tedania (T.) tantula. Data and illustrations of some rare or uncommon species are provided: Iophon gaussi, Lissodendoryx (E.) anacantha, Microxina charcoti, Raspailia (H.) hentscheli, Haliclona spongiosissima, Haliclona (G.) cf. cucurbitiformis. Although the present results increase substantially the previously known information on sponge species, this is still an underestimation of total richness because certain orders (Tetractinellida, Suberitida, Biemnida, Polymastiida) and classes were not studied during the current investigation, although previous records were included.

A lectin from the marine sponge Haliclona (Reniera) implexiformis (HiL) was isolated by affinity chromatography on Sepharose™ matrix. HiL showed specificity for galactose and its derivatives. The glycoproteins porcine stomach mucin (PSM) and bovine stomach mucin (BSM) were potent inhibitors. Hemagglutinating activity of the lectin was maximal between pH 5.0 and 9.0. The lectin remained active until 60°C. The presence of CaCl2 and EDTA did not affect the hemagglutinating activity. In SDS-PAGE, HiL showed a single band of 20 kDa under reduced conditions, whereas in the non-reducing conditions, it showed a band of 20 kDa and one additional band of 36 kDa. The average molecular mass determined by Electrospray Ionization Mass Spectrometry (ESI-MS) was 35.874 ± 2 Da in native and non-reducing conditions, whereas carboxyamidomethylated-lectin showed 18,111 Da. These data indicated that HiL consists in a dimer formed by identical subunits linked by disulfide bonds. Partial amino acid sequence of HiL was determined by mass spectrometry, and revealed that it is a new type of lectin, which showed no similarity with any protein. Secondary structure consisted of 6% α-helice, 31% ß-sheet, 18% ß-turn and 45% random coil. HiL showed significant reduction in the number of viable cells of Staphylococcus biofilms.

A new amide, baeriamide (1), along with nine known diketopiperazines (2-10), was isolated from the marine sponge Haliclona baeri. Their structures were identified by the means of UV, IR, MS and NMR. The absolute configuration of 1 was established by Marfey's method and comparing the specific optical rotation with the known compound HCO-Val-Gly methyl ester. Compound 1 was derived from dehydration of formylated L-valine with γ-amino-butanoic acid methyl ester. Compounds 2-10 were isolated from the genus of Haliclona for the first time. The absolute confirmation of 7 was confirmed first by the means of single-crystal X-ray diffraction. The cytotoxic, antibacterial, antiviral and antifouling activities of these compounds were also tested. However, none of them exhibited significant bioactivities.

In this study, twenty-four organic extracts from six marine sponge species, collected at shallows of Yucatan, Mexico, were evaluated against Giardia lamblia trophozoites and Vero cells. The dichloromethane and hexane extracts of Haliclona tubifera exhibited the highest antigiardiasic activity (IC50 = 1.00 and 2.11 µg/mL, respectively), as well as high selectivity (SI = 41.8 and > 47.4, respectively), while ethyl acetate and methanol extracts of Cinachyrella alloclada, and methanol extract of Suberites aurantiaca showed moderate activity. Contrastingly, the extracts of Halichondria magniculosa and Oceanapia nodosa were considered non actives. Consequently, the dichloromethane extract of H. tubifera were subject to an exploratory chemical study, isolating cholesterol, two benzaldehyde derivatives, three benzoic acid derivatives, cytosine, and thymine.

In this paper, we identify some sponge specimens collected in the Faro Lake in Sicily, and belonging to Haliclona (Halicoclona) by using morphological analysis accompanied by molecular analysis through amplification of several molecular markers (18S and 28S rRNA, CO1 and ITS). The samples are identified as. H. (Halichoclona) vansoesti de Weerdt, de Kluijver & Gómez, 1999, a species native to the Caribbean, and therefore this is the first record of an alien species of the Demospongiae class (Porifera) from the Mediterranean Sea. This presence can be ascribed as results of global change (mainly global warming) that are affecting marine environment.

RESUMO

Twenty-five new species of shallow-water sponges are described from the Galpagos Islands, a province of Ecuador in the eastern Pacific Ocean. Sponges were photographed in situ and collected by SCUBA divers between 2001 and 2004. New species include: Acanthancora equiformis sp. nov., Acanthella saladinorum sp. nov., Cacospongia hermanorum sp. nov., Cinachyrella solis sp. nov., Ciocalypta bustamanti sp. nov., Clathria (Microciona) stellata sp. nov., Clathrina andreusi sp. nov., Craniella lissi sp. nov., Dragmacidon raeae sp. nov., Dragmacidon hendersoni sp. nov., Haliclona (Haliclona) clairae sp. nov., Haliclona (Haliclona) dianae sp. nov., Haliclona (Reniera) oberi sp. nov., Haliclona (Soestella) spuma sp. nov., Haliclona (Soestella) roslynae sp. nov., Hemimycale harlequinus sp. nov., Hemimycale nathani sp. nov., Higginsia johannae sp. nov., Neopetrosia eructans sp. nov., Leucilla agitata sp. nov., Penares angeli sp. nov., Prosuberites vansoesti sp. nov., Suberea esmerelda sp. nov., Tethya annona sp. nov. and Tethya sorbetus sp. nov. In addition, three species have been recorded for the first time from the Galpagos Islands: Chalinula cf. molitba (De Laubenfels, 1949), Chelonaplysilla violacea (Von Lendenfeld, 1883) and Tedania (Tedania) tropicalis Aguilar-Camacho, Carballo Cruz-Barraza, 2018. This paper represents a significant contribution to our knowledge of the Porifera of the Galpagos Islands.

Until now, 127 species of marine sponges have been recorded in the southern Gulf of Mexico (SGoM). In this study, we describe the sponge fauna recorded on 16 coral reefs of the SGoM, defined as the Mexican waters of the Gulf of Mexico (GoM), during a period from 2005 to 2019. We report 80 sponge species, including 34 first geographic records for the southern GoM region. The latter are fully described and illustrated, taking into account 24 that represent new records for the GoM: Agelas conifera, Agelas sventres, Agelas wiedenmayeri, Prosuberites carriebowensis, Desmanthus meandroides, Cliona aprica, Cliona dioryssa, Placospongia ruetzleri, Haliclona (Gellius) megasclera, Haliclona (Reniera) aff. portroyalensis, Neopetrosia proxima, Xestospongia arenosa, Calyx podatypa, Shiphonodictyon xamaycaense, Acarnus innominatus, Iotrochota arenosa, Polymastia tenax, Svenzea cristinae, Svenzea flava, Svenzea tubulosa, Svenzea zeai, Timea stenosclera, Stellettinopsis megastylifera, Suberea flavolivescens. The present work highlights the understimated and remarkable diversity of reef-associated sponges within the Campeche Bank Coral reef systems. Present work data was compiled with existing published information to produce an updated list of 161 known sponges in the southern GoM.

Using bio-guided fractionation and based on the inhibitory activities of nitric oxide (NO) and prostaglandin E2 (PGE2), eight isoquinolinequinone derivatives (1-8) were isolated from the marine sponge Haliclona sp. Among these, methyl O-demethylrenierate (1) is a noble ester, whereas compounds 2 and 3 are new O-demethyl derivatives of known isoquinolinequinones. Compound 8 was assigned as a new 21-dehydroxyrenieramycin F. Anti-inflammatory activities of the isolated compounds were tested in a co-culture system of human epithelial Caco-2 and THP-1 macrophages. The isolated derivatives showed variable activities. O-demethyl renierone (5) showed the highest activity, while 3 and 7 showed moderate activities. These bioactive isoquinolinequinones inhibited lipopolysaccharide and interferon gamma-induced production of NO and PGE2. Expression of inducible nitric oxide synthase, cyclooxygenase-2, and the phosphorylation of MAPKs were down-regulated in response to the inhibition of NF-κB nuclear translocation. In addition, nuclear translocation was markedly promoted with a subsequent increase in the expression of HO-1. Structure-activity relationship studies showed that the hydroxyl group in 3 and 5, and the N-formyl group in 7 may be key functional groups responsible for their anti-inflammatory activities. These findings suggest the potential use of Haliclona sp. and its metabolites as pharmaceuticals treating inflammation-related diseases including inflammatory bowel disease.

Two new marine sponges, Haliclona (Reniera) oceanus sp. nov. and Haliclona (Reniera) juckdoensis sp. nov. of the family Chalinidae were collected from Ieodo Ocean Research Station, Ulleung-do Island, Korea by SCUBA in 2016-2017. Morphologically distinct from each other, both species are assigned to the genus Haliclona subgenus Reniera. Haliclona (Reniera) oceanus sp. nov. is similar to H. (H.) ieoensis Kim et al. 2017 in shape, habitat, growth form, but it differs in color, ectosomal, choanosomal skeleton and spicule size. Haliclona (Reniera) juckdoensis sp. nov. is similar to H. (R.) hongdoensis Kang and Sim 2007 in habitat, ectosomal, choansomal skeleton but it differs in spicule size.

Natural secondary metabolites of sponges of the genus Haliclona are associated with an array of biological activity with therapeutic usage. We investigated the immunopharmacological properties of a presumably novel marine sponge species from Sri Lanka, Haliclona (Soestella) sp. Sponge material was collected from southern Sri Lanka by scuba diving. Sponge identification was based on spicule and skeleton morphology using light microscopy. Selected in vivo and ex vivo tests investigated nonfunctional and functional immunomodulatory activity of the Haliclona (Soestella) sp. crude extract (HSCE) in the Wistar rat model. Compared to the controls, rats orally gavaged daily for 14 consecutive days with 15 mg/kg dose of the HSCE manifested a significant reduction of immune cell counts of total WBCs (by 17%; p < 0.01), lymphocytes (38%), platelets (52%), splenocytes (20%), and bone marrow cells (BMC; 60%) (p < 0.001), with a concurrent increase in the neutrophil : lymphocyte ratio (p < 0.05); RBC counts abated by 53% (p < 0.001). A significant reduction of the splenosomatic index was evident with the 10 and 15 mg/kg doses (p < 0.001). Rat plasma TNF-α cytokine level was augmented by tenfold (p < 0.001), IL-6 level by twofold (p < 0.01) with the 15 mg/kg HSCE treatment, while IL-10 was detectable in rat plasma only with this treatment; the corresponding Th1 : Th2 cytokine ratio (TNF-α : IL-10) was indicative of an unequivocal Th1-skewed cytokine response (p < 0.01). Ex vivo bone marrow cell and splenocyte proliferation were significantly and dose dependently impaired by HSCE (IC50 0.719 and 0.931 µg/mL, respectively; p < 0.05). Subacute toxicity testing established that HSCE was devoid of general toxic, hepatotoxic, and nephrotoxic effects. In conclusion, HSCE was orally active, nontoxic, and effectively suppressed nonfunctional and functional immunological parameters of Wistar rats, suggestive of the potential use of the HSCE as an immunosuppressant drug lead.

Haliclona (Flagellia) Van Soest, 2017 is a recently erected subgenus characterized by the presence of flagellosigma microscleres which are often distinctive between species (Van Soest 2017). Members of the taxon also have normal sigmas within a confused skeleton formed by oxea megascleres. The subgenus has a global distribution and contains 10 species (Van Soest et al. 2019). A large and abundant new species collected throughout the southern Gulf of St. Lawrence is characterized by thick and abnormally shaped flagellosigmas, two sizes of oxea, and abundant normal sigmas. Lambe (1896) previously reported Haliclona (Flagellia) porosa (Fristedt, 1887) from the Gulf of St. Lawrence, and the species is known to occur in nearby Arctic waters (Fristedt 1887; Lundbeck 1902; Hentschel 1916; Koltun 1959, 1966; Van Soest 2017; Dinn Leys 2018). However, H. (F.) porosa is characterized by the presence of very few normal sigmas and no thick flagellosigmas. Type specimens were preserved in 95% ethanol and were deposited in the Atlantic Reference Centre (ARC) in St. Andrews, New Brunswick, Canada. The taxonomic identification was performed through spicule analysis using light and scanning electron microscopy, following methods described by Dinn et al. (2020).

Porifera has been relatively well studied from underwater caves worldwide. However, sponges in Mexico are only known from two anchialine caves: La Quebrada and El Aerolito, both in Cozumel Island. An ecological study found that sponges have the second highest species richness and density in La Quebrada. The aim of the present work is to describe the sponge biodiversity inhabiting the dark zone of La Quebrada anchialine cave. A total of ten species were found and are described herein, six are new species to science vis. Calyx maya n. sp. a thinly branched sponge; Haliclona (Reniera) stygobia n. sp. globe-shaped with the thinnest diameter of oxeas in its group; Haliclona (Halichoclona) chankanaabiis n. sp. tubular habit with the smallest oxeas in its group; Neosiphonia microtriaeneae n. sp. characterized by very small dichotriaenes; Svenzea germanyanezi n. sp. a lone conical shape; and Diplastrella cozumella n. sp. for its tri to multilobed tylostyles. The other four species are previously known from open reefs or deep-sea: Discodermia adhaerens, Siphonidium ramosum, Cinachyrella kuekenthali, and Plakinastrella onkodes. This is the only inventory of Porifera in Mexican caves up to now, thus the species here described are the first stygobiont sponges from Mexico with an endemism rate of Porifera in this anchialine cave of 60%. Conservation programs are needed to preserve this unique and special ecosystem.

Seven new polyoxygenated steroids belonging to a new structural group of sponge steroids, gracilosulfates A-G (1-7), possessing 3ß-O-sulfonato, 5ß,6ß epoxy (or 5(6)-dehydro), and 4ß,23-dihydroxy substitution patterns as a common structural motif, were isolated from the marine sponge Haliclona gracilis. Their structures were determined by NMR and MS methods. The compounds 1, 2, 4, 6, and 7 inhibited the expression of prostate-specific antigen (PSA) in 22Rv1 tumor cells.

Chemical study of the CH2Cl2-MeOH (1:1) extract from the sponge Haliclona sp. collected in Mayotte highlighted three new long-chain highly oxygenated polyacetylenes, osirisynes G-I (1-3) together with the known osirisynes A (4), B (5), and E (6). Their structures were elucidated by 1D and 2D NMR spectra and HRESIMS and MS/MS data. All compounds were evaluated on catalase and sirtuin 1 activation and on CDK7, proteasome, Fyn kinase, tyrosinase, and elastase inhibition. Five compounds (1; 3-6) inhibited proteasome kinase and two compounds (5-6) inhibited CDK7 and Fyn kinase. Osirisyne B (5) was the most active compound with IC50 on FYNB kinase, CDK7 kinase, and proteasome inhibition of 18.44 µM, 9.13 µM, and 0.26 µM, respectively.

An automated, high-capacity, and high-throughput procedure for the rapid isolation and identification of biologically active natural products from a prefractionated library is presented. The semipreparative HPLC method uses 1 mg of the primary hit fraction and produces 22 subfractions in an assay-ready format. Following screening, all active fractions are analyzed by NMR, LCMS, and FTIR, and the active principle structural classes are elucidated. In the proof-of-concept study, we show the processes involved in generating the subfractions, the throughput of the structural elucidation work, as well as the ability to rapidly isolate and identify new and biologically active natural products. Overall, the rapid second-stage purification conserves extract mass, requires much less chemist time, and introduces knowledge of structure early in the isolation workflow.

Halococcus agarilyticus GUGFAWS-3 (MF425611) was isolated from a marine white sponge of Haliclona sp., inhabiting the rocks in the intertidal region of Anjuna, Goa, India. Uniquely, the microbe simultaneously produces two halo-extremozymes in 25% NaCl, namely protease and lipase at 49.5 ± 0.4 and 3.67 ± 0.02 (U mL-1), respectively. The protease is constitutively produced in starch mineral salts medium with consistent 4 ± 1.0 mm zone of enzyme production, regardless of the non-availability of protein as substrate. The ethanol precipitated enzyme on dialysis and Sephadex G-200 gel filtration chromatography was partially purified to 12.26-fold and was active between 20 and 80 °C, 0-5 M NaCl, and pH 3-13. Optimum activity, however, was at 70 °C, 3 M NaCl, and pH 7. The enzyme was thermo stable at 70 °C with 50.26 ± 2.40% of relative enzyme activity at 75 min. Furthermore, it was stable in the presence of polar and non-polar organic solvents, detergents, and hydrocarbons. Several metal cations enhanced its activity in the order of Ca2+ > Ni2+ > Fe3+ > Co2+ > Mg2+ > Cu2+ > Mn2+. Dependence of enzyme on cysteine; serine, and metal ions was confirmed by ß-mercaptoethanol; PMSF and EDTA, respectively which induced its partial inhibition. Additionally, protease inhibited in vitro biofilm formation in Staphylococcus aureus. Conclusively, the production of a neutral halo-thermophilic protease is reported for the first time in the genus Halococcus.

RESUMO

Marine sponge genus Haliclona, one of the most prolific sources of natural products, contains over 600 species but only a small part of them had been classified and chemically investigated. On the basis of extensive literature search, this review firstly summarizes 112 nitrogenous secondary metabolites from classified and unclassified Haliclona sponges as well as from their symbiotic microorganisms. Most of these substances have only been found in Haliclona sponges, and display diverse bioactive properties with potential applications in new drug discovery.

Three new sponge species in the demosponge families Chalinidae Gray and Suberitidae Schmidt are described from the Calypso hydrothermal vent field in the Bay of Plenty, and one species from seep sites along the Hikurangi Margin, to the east of the North Island, New Zealand. The Calypso hydrothermal vent field is dominated by the chalinid sponge Haliclona (Soestella) battershilli sp. nov., a large, cream-coloured, finely branched species, and the less common H. (Halichoclona) sonnae sp. nov., an encrusting, translucent white, cushion-shaped species. The third species, the suberitid sponge Protosuberites novaezelandiae sp. nov., forms encrustations with digitate projections. Haliclona (Halichoclona) sonnae sp. nov. and Protosuberites novaezelandiae sp. nov. represent new subgenus and genus records, respectively, for New Zealand waters. The methanotrophic suberitid sponge, Pseudosuberites thurberi sp. nov., is found at many of the cold seep sites on the Hikurangi Margin where it forms extensive, encrusting to digitate mats. The description of these species provides a basis for the future study of the ecology of sponges that are apparently endemic to vent and seep habitats off New Zealand.

Marine sponges often house small-molecule-producing symbionts extracellularly in their mesohyl, providing the host with a means of chemical defence against predation and microbial infection. Here, we report an intriguing case of chemically mediated symbiosis between the renieramycin-containing sponge Haliclona sp. and its herein discovered renieramycin-producing symbiont Candidatus Endohaliclona renieramycinifaciens. Remarkably, Ca. E. renieramycinifaciens has undergone extreme genome reduction where it has lost almost all necessary elements for free living while maintaining a complex, multi-copy plasmid-encoded biosynthetic gene cluster for renieramycin biosynthesis. In return, the sponge houses Ca. E. renieramycinifaciens in previously uncharacterized cellular reservoirs (chemobacteriocytes), where it can acquire nutrients from the host and avoid bacterial competition. This relationship is highly specific to a single clade of Haliclona sponges. Our study reveals intracellular symbionts as an understudied source for defence chemicals in the oldest-living metazoans and paves the way towards discovering similar systems in other marine sponges.

Salt stable cellulases are implicated in detritic food webs of marine invertebrates for their role in the degradation of cellulosic material. A haloarchaeon, Haloferax sulfurifontis GUMFAZ2 producing cellulase was successfully isolated from marine Haliclona sp., a sponge inhabiting the rocky intertidal region of Anjuna, Goa. The culture produced extracellular xylanase-free cellulase with a maximum activity of 11.7 U/ml, using carboxymethylcellulose-Na (CMC-Na), as a sole source of carbon in 3.5 M NaCl containing medium, pH 7 at 40°C and produced cellobiose and glucose, detectable by thin-layer chromatography. Nondenaturing polyacrylamide gel electrophoresis of the crude enzyme, revealed a single protein band of 19.6 kDa which on zymographic analysis exhibited cellulase activity while corresponding sodium dodecyl sulfate polyacrylamide gel electrophoresis revealed a molecular weight of 46 kDa. Unlike conventional cellulases, this enzyme is active in presence of 5 M NaCl and does not have accompanying xylanase activity, hence can be considered as xylanase-free cellulase. Such enzymes from haloarchaea offer great potential for biotechnological application because of their stability at high salinity and is therefore worth pursuing.
